چکیده

Background The present work evaluates the cross-reactivity of commercially available cyclic nucleotide analogs with cAMPand cGMP-immunoassays from Cayman, IBL (both IBL International, Hamburg, Germany) and ENZO Life Sciences (Loerrach, Germany). Results and conclusion Most of the tested cyclic nucleotide analogs showed low degree competition with the antibodies; however, with Rp-cAMPS, 8-Br-cGMP and 8-pCPT-cGMP a strong cross-reactivity with the ENZO cAMPrespectively cGMP-EIA and the IBL cGMP-RIA was observed (Table

CYCLIC NUCLEOTIDES CONTROL DIFFERENTIATION OF HUMAN MONOCYTES INTO EITHER HIGHLY ACCESSORY CELLS OR MACROPHAGES

Human peripheral blood monocytes have been found to undergo a transitory state of high accessory activity before they fully become macrophages. Time kinetics were done to follow this accessory potential. Studying the regulation of accessory activity, we have found that monocyte derived accessory cells (m-AC) pass through two phases of development, both of which are adversely controlled by ...

Introduction of Aromatic Ring-Containing Substituents in Cyclic Nucleotides Is Associated with Inhibition of Toxin Uptake by the Hepatocyte Transporters OATP 1B1 and 1B3

Analogs of the cyclic nucleotides cAMP and cGMP have been extensively used to mimic or modulate cellular events mediated by protein kinase A (PKA), Exchange protein directly activated by cAMP (Epac), or protein kinase G (PKG). We report here that some of the most commonly used cyclic nucleotide analogs inhibit transmembrane transport mediated by the liver specific organic anion transporter pept...

Expression of Recombinant Phosphodiesterase 3A and 3B Using Baculovirus Expression System

Background: Phosphodiesterase 3A (PDE3A) and phosphodiesterase 3B (PDE3B) play a critical role in the regulation of intracellular level of adenosine 3´,5´-cyclic monophosphate (cyclic AMP, cAMP) and guanosine 3´,5´-cyclic monophosphate (cyclic GMP, cGMP). Subsequently PDE3 inhibitors have shown to relax vascular and inhibit platelet aggregation in cardiovascular disease. Objectives: In th...

"cAMP-specific" phosphodiesterase contributes to cGMP degradation in cerebellar cells exposed to nitric oxide.

Nitric oxide (NO) functions as a diffusible messenger in the central nervous system and elsewhere, exerting many of it physiological effects by activating soluble guanylyl cyclase, so increasing cellular cGMP levels. Hydrolysis of cyclic nucleotides is achieved by phosphodiesterases (PDEs) but the enzyme isoforms responsible for degrading cGMP in most cells have not been identified. We have dev...

Cyclic nucleotide modulation of in vitro morphological transformation of Syrian hamster cells.

The cyclic nucleotides, cyclic adenosine 3':5'-monophosphate (cAMP) and cyclic guanosine 3':5'-monophosphate (cGMP) or their dibutyryl and monobrominated derivatives, may either increase or decrease morphological transformation of Syrian hamster embryo cells exposed to N-methyl-N'-nitro-N'-nitrosoguanidine (MNNG). The effect on transformation is primarily a function of the parent cyclic nucleot...
